I went to update one of the digital signage systems using the Content Manager Desktop for FWI and received that error. I think a recent update to the player software from 5 to 5.3 may have been what caused it. Here's what I did to fix it:

FWI cannot deploy to any of the player machines
Four Winds Interactive Error: Cannot deploy to any of the player machines. Check your network configuration or machine properties and deploy again. Status: Player machine not configured.
